Determination of Reaction Kinetic Parameters for Inactivation of Enzymes Usinga Differential Scanning Calorimeter

Abstract

The reaction kinetic parameters were determined using differential scanning calorimetery (DSC) for non-isothermal inactivation of a lipoxygenase and a peroxidase. From the DSC thermogram containing the come-up and the cooling time, the D value and z value for the lipoxygenase were 22.0 min and 7.1¡É, respectively. Both the activation energy (E_a) and the reaction rate constant (k) as well as other kinetic parameters were relatively in good agreement with the results previously reported on the lipoxygenase since it was inactivated as first-order kinetics. However, the results obtained from the peroxidase by DSC showed considerable discrepancy with the kinetic parameters of isothermal inactivation. The reaction order of the peroxidase from non-isothermal inactivation using DSC was 1.3 while that obtained from isothermal treatment was 1.7. The results strongly supported that the kinetic parameters obtained through DSC could provide clearer explanations for the thermal inactivation behavior.
